Microspectroscopy of single proliferating HeLa cells

چکیده
Spectral differences between normal and abnormal tissue observed to date appear to be due to different averaging processes of spectral patterns that differ according to the cell’s biochemistry, due to its state of maturation, differentiation, and development. Thus, disease perturbs the distribution of cells in the different stages of maturation, differentiation, and development. Previous FTIR microspectroscopic studies of normal versus neoplastic cells and tissues have demonstrated differences in the absorption intensities and band-shapes, particularly in the low frequency (1200–1000 cm ) spectral region. In this study, we further investigated the spectral changes due to the drastic biochemical and morphological changes occurring as a consequence of cell proliferation. # 2004 Elsevier B.V. Cellular injury evidenced by impedance technology and infrared microspectroscopy.
Fourier Transform Infrared (FTIR) spectroscopy is finding increasing biological application, for example in the analysis of diseased tissues and cells, cell cycle studies and investigating the mechanisms of action of anticancer drugs. Cancer treatment studies routinely define the types of cell-drug responses as either total cell destruction by the drug (all cells die), moderate damage (cell det...
متن کاملDNA Methylation Changes in Valproic Acid-Treated HeLa Cells as Assessed by Image Analysis, Immunofluorescence and Vibrational Microspectroscopy
Valproic acid (VPA), a well-known histone deacetylase inhibitor, has been reported to affect the DNA methylation status in addition to inducing histone hyperacetylation in several cell types. In HeLa cells, VPA promotes histone acetylation and chromatin remodeling. However, DNA demethylation was not checked in this cell model for standing effects longer than those provided by histone acetylatio...
متن کاملProliferating Pilomatricoma: A Case Report and Review of Literature
Proliferating pilomatricoma is a distinctive rare variant of pilomatricoma first described in 1997.Very few cases of proliferating pilomatricoma have been reported in the literature until date. This report is a case of 65 years old male presenting with hard nodular lump on the right side of neck slowly increasing in size for the last 8 months. Wide excision of the lump was performed with clear ...
متن کاملIdentification of S-phase Cells with PCIO Antibody to Proliferating Cell Nuclear Antigen
We estimated the expression of proliferating cell nuclear antigen (PCNA) in HeLa S3 cells by flow cytometry with monoclonal antibody (MAb) PC10. HeLa cells were fued with six Merent fuation procedures: 15-mk and 30-min acetone, 15-mb acetone followed by 15-mb methanol (acetonelmethanol), 30-mb methanol, 15-min methanol followed by 15-mh acetone (methanollacetone), and a mixture of acetone and m...
متن کاملHuman APE2 protein is mostly localized in the nuclei and to some extent in the mitochondria, while nuclear APE2 is partly associated with proliferating cell nuclear antigen.
In human cells APE1 is the major AP endonuclease and it has been reported to have no functional mitochondrial targeting sequence (MTS). We found that APE2 protein possesses a putative MTS. When its N-terminal 15 amino acid residues were fused to the N-terminus of green fluorescent protein and transiently expressed in HeLa cells the fusion protein was localized in the mitochondria. By electron m...
